The layout I built now almost 30 years ago was done with Homosote. I will soon be building an extension for a staging yard. It will be built using the same method and materials. I am also building a small N scale layout and will use Homosote on it as well. It is sound deadening and holds track nails well. Track can be rearranged with ease. Scenery can be glued to it with ease. I have never had an issue with humidity or dampness. I cut it outside so dust is not a problem. If for some reason it must be cut inside, I use a jigsaw knife blade. This eliminates the issue of dust.
I use blue or pink foam for scenic land forms. It is easily cut and shaped. It will melt if exposed to petroleum based paint and glue. It also melts when exposed to heat and produces an acrid odor.

Perhaps I missed it in the flurry of email on this thread, but I’ve not seen mention of sound. My limited experience is that trains are noisy running on foam. Perhaps scenery and other coverings will help diminish the apparent amplification properties of rigid foam.

Homosote is becoming a rarity in use for layouts. It is scarce and hard to locate. Either pink or blue foam board is preferred, especially when using a 1/4" plywood base. The nice thing is that you can use foam board glue applied with a caulking gun to build your base and scenery. 1" foam board is more versatile to use and easily cut to build your scenery. You can laminate different thickness to get to the height you want.
